<image:caption>GLADYS AND ROLAND HARRIMAN PROFESSOR OF IMMUNOLOGY Investigates the mechanisms of innate and adaptive immune responses particularly to microbial infection and cell death. Our laboratory investigates the core mechanisms of innate immunity and inflammation, elucidating their critical roles in human health and disease. We focus on the functions of phagocytes—key immune cells—in combating infections, malignancies, and chronic inflammatory states. Through this research, we aim to drive the development of novel therapeutic strategies and vaccines to address pressing medical challenges. The Blander laboratory is located at Weill Cornell Medicine, within The Jill Roberts Institute for Research in Inflammatory Bowel Disease The Department of Medicine The Department of Microbiology and Immunology The Sandra and Edward Meyer Cancer Center The Cornell Center for Immunology The Weill Cornell/Sloan Kettering Institute Immunology and Microbial Pathogenesis Ph.D. Graduate Program</image:caption>

      <image:title>Research Topic 2</image:title>
<image:caption>The vita-PAMP bacterial mRNA drives augmented follicular T helper cell and antibody responses. Detection of the vita-PAMP bacterial mRNA by patrolling monocytes in the context of Gram-negative bacteria triggers TRIF signaling and IRF3-dependent interferon (IFN) beta production, which engages IFNAR to license activation of the inflammasome and lead to the production of the alert cytokine interleukin-1 (IL-1) beta. IFN-beta and IL-1 beta engage their respective receptors on CD4 T cells instructing their differentiation into follicular T helper cells (TFH), known for their roles in helping B cells make antibodies. This orchestrated series of events from detection of a vita-PAMP to the adaptive humoral immune response illustrates how key first events in innate immunity shape the quality of the adaptive immune response. Whole mount of small intestine in this mouse model stained with antibody to CD11c (white) to visualize the intestinal lamina propria mononuclear phagocytes and antibody to cleaved Caspase-3, an effector of apoptosis (red) to visualize apoptotic intestinal epithelial cells. The model is engineered to express a fusion protein of GFP (not shown) and diphtheria toxin receptor driven by the intestinal epithelium-specific villin-promoter. Intraperitoneal injection of diphtheria toxin elicits apoptosis of diphtheria toxin receptor expressing intestinal epithelial cells. See Cummings R et al. Nature 2016.</image:caption>
